Human serum albumin (HSA)-coated silica nanoparticles (NPs) were fabricated for the first time by the reaction between amino groups of HSA and 3-(trimethoxysilyl) propyl methacrylate. The size, morphology and inner structures of the HSA-coated NPs were characterized by high-resolution transmission electron microscopy, zeta potential and Fourier transform infrared spectroscopy. The morphology of the HSA-coated NPs was spherical and their sizes were mainly in the range of 20-40 nm. The amount of amino groups on the surface of the HSA-coated NPs could be controlled by the amount of 3-(trimethoxysilyl) propyl methacrylate reacted with HSA. The immobilization of anti-CEA onto the HSA-coated NPs was investigated by electrochemical impedance spectroscopy and cyclic voltammetry. The diameter of the anti-CEA immobilized on the HSA-coated NPs was found to be about 7 nm, which was measured by atomic force microscopy. The electrochemical impedance spectroscopy and cyclic voltammetry results revealed that the electrochemical behaviors of the anti-CEA immobilized on the HSA-coated NPs were similar to the free anti-CEA.
